ABSTRACT

ObjectiveTo present a method for evaluating the gait symmetry of microprocessor-controlled prosthetic knee (MPK). MethodsA kind of proto-MPK, AiKneeOne, and a wearable gait collect system, were made. The phases of the first double-limb support, the single-limb support, the second double-limb support, and the swing were used to calculate symmetry index (SI), ratio Ⅰ (RI) and ratio II (RII). Five heathy persons walked on the treadmill wearing AiKneeOne at speeds of 0.5, 0.7, 1.1 m/s, and the indice were collected with the wearable gait collect system. ResultsUnder different velocities, The absolute value of SI and RII were very little and the RI were close to one at the phases of the first double-limb support and the second double-limb support, but they were not very satisfactory in the phases of the single-limb support and the swing. ConclusionThe developed MPK AiKneeOne is potential to reconstruct the gait of amputees, and the gait symmetry indice can be used to evaluate the wearing performance of MPK.

ABSTRACT

Objective:To evaluate the treatment of infected nonunion after internal fixation of subtrochanteric fracture with a reconstruction stent of external fixation.Methods:A retrospective study was conducted to analyze the data of 5 male patients with infected nonunion after internal fixation of subtrochanteric fracture who had been treated and completely followed up at The Great Wall Orthopaedics and Hand Surgery Hospital from January 2017 to October 2022. The patients were (30.0±13.5) years old. Seinsheimer fracture types: ⅢA (1 case), ⅢB (1 case), Ⅳ (2 cases), and Ⅴ (1 case); original internal fixation: intramedullary system (4 cases) and plate fixation (1 case); the Cierny-Mader anatomical classification: type Ⅳ (diffuse type) for all. After complete debridement at stage one, 2 or 3 hydroxyapatite (HA) coated screws were placed at both fracture ends from the lateral side of the femur for unilateral reconstruction external fixation. Next, a hybrid external fixation scaffold was added with a 1/3 ring at the sagittal position and 1 or 2 HA screws in 4 cases while unilateral reconstruction external fixation was constructed at both sides by inserting 2 HA screws into both fracture ends from the anterior femur at the sagittal position in 1 case. Antibiotic bone cement was used to fill bone defects of (3.8±1.8) cm. At 6 to 8 weeks after debridement when infection did not recur, antibiotic bone cement was removed before autogenous iliac bone grafting was performed in 3 patients and osteotomy bone transport in 2 patients. Infection control, bone union time, time for removal of external fixation stent, complications, Sanders hip function score and Paley bone outcome score were recorded.Results:The 5 patients were followed up for (23.4±8.1) months after surgery. Infection at the fracture ends was controlled after 1 time of debridement in 3 patients and after 2 times of debridement in 2 patients. The loosening HA screws were replaced twice due to infection at the proximal nail tract, and autologous bone grafting was performed at the opposite fracture ends in 1 case; no complications occurred in the other 4 cases. Bony union was achieved at the extended segment and fracture ends in all patients. The time for imaging union after bone reconstruction was (10.2±3.4) months. The time for wearing a stent of external fixation was (18.0±4.5) months. There was no recurrent infection or lingering infection. According to the Sanders hip function score at the last follow-up, 4 cases were excellent and 1 case was good; according to the Paley bone outcome score, the curative effect was excellent in all.Conclusion:Application of a reconstruction stent of external fixation combined with antibiotic bone cement can control infection at the first stage and conduct bone reconstruction at the second stage to successfully treat the infected nonunion and preserve the hip function after internal fixation of subtrochanteric fracture.

ABSTRACT

Objective:To explore the application and effect of flipped classroom in community clinical pharmacist training.Methods:Seventeen pharmacists trained in 2017-2018 were used as the control group and received traditional teaching mode. Fourteen pharmacists trained in 2019-2020 were used as the experimental group and received the flipped classroom teaching mode. Finally, the teaching effect was evaluated through the trainees' graduation examination results and satisfaction degree. The SPSS 25.0 was used to conduct the t test and chi-square test. Results:The task performance, prescription audit scores, clinical practice scores, and total scores in the experimental group were significantly higher than those in the control group ( P<0.05). However, there was no significant difference in usual performance between the two groups ( P>0.05). For satisfaction survey, the total score of Likert scale in the experimental group was (20.1±3.4), which was significantly higher than that in the control group (16.9±3.4). However, there was no significant difference in the satisfaction of clinical practice and teachers' level between the two groups ( P>0.05). Conclusion:Flipped classroom can improve the enthusiasm of community clinical pharmacist trainees to participate in learning, increase the level of the trainees' theoretical knowledge and professional practice, and is conducive to the cultivation of high-quality community clinical pharmacists.

ABSTRACT

Objective:To summarise the advantages and disadvantages of applying the method of retrograde replantation with an operative position of foot stepping and knee bending for replantation of rotational avulsed and fractured great toes.Methods:From January 2016 to June 2021, 11 rotational avulsed and fractured great toes were replanted with the method of retrograde replantation in an operative position of foot stepping and knee bending. Of the 11 patients, 10 were males and 1 was female, aged 18 to 50 years old with an average of 32 years old. Causes of injury: 5 of driving belt, 4 of machinery crush and 2 of car accident. Injury sites: 4 on left great toes and 7 on right great toes. All of the injuries were with fracture and exposure of proximal tendons of flexor and extensor. Seven patients had the follow-up reviews by outpatient clinic visiting, 2 over mobile phone and 2 via WeChat.Results:After surgery, 9 great toes completely survived and 2 great toes had necrosis. The survival rate of toe replantation was 81.8%(9/11). The operation time was 2 to 3 hours, with an average of 2.5 hours. Postoperative X-ray film showed that 8 to 12 weeks after the operation, the fracture and joint fusion were healed at first stage in the survived toes. All patients were entered in follow-up for 3 to 18 months with 10.5 months in average. The survived great toes were plump and the toenail grown well. At the final follow-up, the static TPD at the toes was 8-12 mm, with an average of 10 mm. There was no effect shown on either walking or running.Conclusion:The retrograde replantation method with an operative position of foot stepping and knee bending for great toe replantation has the characteristics of a good field of view and convenient in operation under microscope, a short operation time, and a high survival rate of replantation.

Objective:Using the information technology of knowledge element theory to analyze the content of the ancient Chinese medicine book Nv Ke Xin Fa, and to explore the original thinking and implicit knowledge of the author of the book Nv Ke Xin Fa. Methods:Based on the theory and technology of knowledge element, the indexing data was read as logical data by using MS SQL Server database and then the knowledge graph of traditional Chinese medicine was constructed by using neo4j database.Results:There are 345 knowledge bodies and 1 478 knowledge elements in the Nv Ke Xin Fa, mainly including prescription name (280), composition (275) and usage (224); There are 711 semantic types, mainly including prescriptions (232), etiology and pathogenesis (221), syndrome manifestations (125) and treatment (88); 187 semantic associations, mainly syndrome differentiation and treatment (172); Knowledge of semantic association.There are 34 body types, with leucorrhea (16), amenorrhea (13), blood Avalanche (13) and asthenia (13). Conclusions:Zheng Qinyu paid attention to distinguishing the texture of leucorrhea in the treatment of leucorrhea, put forward the pathogenesis and treatment principles and methods of leucorrhea in the stages of "before childbirth" and "after childbirth", and made it clear that the application scope of Buzhong Yiqi Decoction involves menstruation, leucorrhea, pregnancy and the whole cycle of childbirth.Mrs Zheng Qinyu pays great attention to the protection on the spleen and stomach of the middle coke and regulation of Qi that could rise and fall, which could provide a reference for modern clinical diagnosis and treatment of leukorrhagia diseases.

ABSTRACT

The responsive patterns of phytochrome gene family members to photoperiod and abiotic stresses were comparatively analyzed and the favorable natural variation sites of these genes were identified. This would help understand the mechanism of phytochrome gene family in photoperiod-regulated growth and development and abiotic stress response. In addition, it may facilitate the molecular marker assisted selection of key traits in foxtail millet. In this study, we used RT-PCR to clone three phytochrome genes SiPHYA, SiPHYB and SiPHYC from ultra-late maturity millet landrace variety 'Maosu'. After primary bioinformatics analysis, we studied the photoperiod control mode and the characteristics of these genes in responding to five abiotic stresses including polyethylene glycol (PEG)-simulated drought, natural drought, abscisic acid (ABA), high temperature and NaCl by fluorescence quantitative PCR. Finally, we detected the mutation sites of the three genes among 160 foxtail millet materials and performed haplotype analysis to determine the genes' functional effect. We found that the cloned cDNA sequences of gene SiPHYA, SiPHYB and SiPHYC were 3 981, 3 953 and 3 764 bp respectively, which contained complete coding regions. Gene SiPHYB and SiPHYC showed closer evolutionary relationship. Photoperiod regulated all of the three genes, but showed more profound effects on diurnal expression pattern of SiPHYB, SiPHYC than that of SiPHYA. Under short-day, when near heading, the expression levels of SiPHYA and SiPHYB were significantly lower than that under long-day, indicating their roles in suppressing heading of foxtail millet under long-day. SiPHYB and SiPHYC were responsive to PEG-simulated drought, natural drought, ABA and high temperature stresses together. SiPHYA and SiPHYB responded differently to salt stress, whereas SiPHYC did not respond to salt stress. Re-sequencing of 160 foxtail millet materials revealed that SiPHYB was highly conservative. Two missense mutations of SiPHYA, such as single nucleotide polymorphism (SNP) 7 034 522C→T and SNP7 036 657G→C, led to delaying heading and increasing plant height. One missense mutation of SiPHYC, such as SNP5 414 823G→T, led to shortening heading under short-day and delaying heading under long-day, as well as increasing plant height and panicle length regardless of photo-thermal conditions. Photoperiod showed different regulatory effects on SiPHYA, SiPHYB and SiPHYC. SiPHYB and SiPHYC jointly responded to various abiotic stresses except for the salt stress. Compared with the reference genotype, mutation genotypes of SiPHYA and SiPHYC delayed heading and increased plant height and panicle length.

ABSTRACT

B-cell precursor acute lymphoblastic leukemia (BCP-ALL) is characterized by genetic alterations with high heterogeneity. Precise subtypes with distinct genomic and/or gene expression patterns have been recently revealed using high-throughput sequencing technology. Most of these profiles are associated with recurrent non-overlapping rearrangements or hotspot point mutations that are analogous to the established subtypes, such as DUX4 rearrangements, MEF2D rearrangements, ZNF384/ZNF362 rearrangements, NUTM1 rearrangements, BCL2/MYC and/or BCL6 rearrangements, ETV6-RUNX1-like gene expression, PAX5alt (diverse PAX5 alterations, including rearrangements, intragenic amplifications, or mutations), and hotspot mutations PAX5 (p.Pro80Arg) with biallelic PAX5 alterations, IKZF1 (p.Asn159Tyr), and ZEB2 (p.His1038Arg). These molecular subtypes could be classified by gene expression patterns with RNA-seq technology. Refined molecular classification greatly improved the treatment strategy. Multiagent therapy regimens, including target inhibitors (e.g., imatinib), immunomodulators, monoclonal antibodies, and chimeric antigen receptor T-cell (CAR-T) therapy, are transforming the clinical practice from chemotherapy drugs to personalized medicine in the field of risk-directed disease management. We provide an update on our knowledge of emerging molecular subtypes and therapeutic targets in BCP-ALL.

ABSTRACT

Objective:To report our experience in successful limb salvage in 2 cases of femoral fracture of Gustilo type ⅢC after warm ischemia beyond 10 hours.Methods:From February 2016 to April 2017, 2 patients with femoral fracture of Gustilo type ⅢC were treated at The Great Wall Orthopaedic and Hand Surgery Hospital after warm ischemia beyond 10 hours.Both of them were male, 42 and 27 years of age.After debridement, bone shortening for 8.0 cm and 10.0 cm respectively was followed by replantation. The fractures were fixated end to end with a compression plate, the femoral artery was anastomosed directly and the muscles were sutured end to end.Secondary osteotomy was conducted 4 months after successful replanta-tion, followed by unilateral external fixation for reconstruction.Limb lengthening began one week after sec-ondary osteotomy.The efficacy was assessed by the Paley criteria.Results:Scattered irregular necrosis occurred in the leg muscles but was cured by dermatoplasty through a decompressive incision 40 days after debridement for 4 times.The affected limbs in the 2 cases were successfully saved and lengthened to the same length as the opposite one.The limbs were lengthened for 8.0 and 10.0 cm and for 3.0 and 3.5 months, respectively.The unilateral external fixator was worn for 29 and 24 months, respectively.The plantar sensation recovered to S4 one year after operation.Follow-up at 6 months after removal of external fixation showed excellent bony outcomes and good functional outcomes in both cases According to the Paley criteria.Conclusion:For femoral fracture of Gustilo type ⅢC after warm ischemia beyond 10 hours, thorough debridement, plate fixation, bone shortening and replantation at one stage, followed by secondary limb lengthening using unilateral external fixation, may be practical and effective procedures, as long as in-dications for limb salvage should be comprehensively followed.

ABSTRACT

Objective:To investigate the biocompatible properties of tissue-engineered rabbit trachea treated by Triton-X 100 processed method (TPM) and detergent enzymatic method (DEM) with genipin cross-linking.Methods:TPM and DEM were used to decellularize New Zealand rabbit trachea, and then genipin was used for cross-linking. The mechanical properties of each tracheal sample were measured by universal tensile testing machine. The structure of the sample was observed by scanning electron microscope. The cytotoxicity of the sample was detected by cell contact toxicity assay. Fifteen healthy adult New Zealand rabbits with no specific pathogens were divided into the native tracheal transplantation group, the genipin cross-linked TPM acellular tracheal matrix transplantation group and the genipin cross-linked DEM acellular tracheal matrix transplantation group according to the random number table, 5 animals for each group. Animals in each group were sacrificed 30 days after transplantation, and graft samples were obtained. The microstructure was observed by hematoxylin-eosin staining and CD68 molecular immunohistochemical staining.Results:Biomechanical results showed that the mechanical properties of decellularized tracheas with genipin cross-linking were similar to native tracheas. The results of scanning electron microscopy showed that the matrix of cross-linked decellularized tracheas was more dense comparing with native tracheas, and the mesh-like ultrastructure formed on the outer surface of the genipin cross-linked DEM acellular tracheal matrix was conducive to cell adhesion. The results of cell contact toxicity results showed that the genipin cross-linked decellularized tracheas treated by DEM had better biocompatibility. The results of in vivo implantation and histological staining showed that genipin cross-linked DEM acellular tracheal matrix was less immunogenic comparing with genipin cross-linked TPM acellular tracheal matrix.Conclusions:Genipin can improve the ultrastructure of decellularized tracheal matrix without causing inflammatory. The genipin cross-linked decellularized tracheas treated by DEM has better biocompatibility and lower immunogenicity, which make it suitable for the replacement of tissue engineering trachea.

ABSTRACT

Objective:To investigate the application of low-dose chest CT(LDCT) in the screening of pulmonary subsolid nodules in population and the application value of artificial intelligence.Methods:People who received chest LDCT screening between January 2015 and December 2017 were included. A retrospective study was developed to analyze the enrolled population features , detection of pulmonary subsolid nodules and independent predictors of subsolid nodules , and to evaluate the accuracy of the artificial intelligence reading method.Results:Result of three cross-sectional studies reveals that the detection rates of pulmonary subsolid nodules were 0.42%, 0.69% and 0.92% in three rounds. 726 cases who completed the three rounds of screening were included in the cohort study. The cohort population was predominantly male(83.2%), with a median age of 43 years, and nearly half of the subjects(47.0%) had a history of smoking. GEE revealed that the patient's family history of lung cancer( OR=8.753, 95% CI: 1.877-40.816, P=0.006) was an independent predictor of the detection of subsolid nodules. In the 110 kVp tube voltage group, AUC of AI model was 0.740, and AUC of the manual reading method was 0.721, no significant differences were observed( P=0.502); when the preseted cutoff value of AI model was 0.75, the NRI was -0.15, indicating the accuracy of AI model was inferior to manual method( P=0.006). In the 130 kVp tube voltage group, AUC of the model was 0.888, and AUC of the manual reading method was 0.756, no significant differences were observed( P=0.128); and the NRI was 0.19, indicating the accuracy of AI model was not inferior to manual method( P=0.123). Conclusion:This population' s detection rates of pulmonary subsolid nodules were 0.42%-0.92%. Family history of lung cancer was an independent predictor of subsolid pulmonary nodules. The result of AI pulmonary nodule detection model could be a reference when the training set data parameters match the actual application parameters.

ABSTRACT

Objective:To explore the clinical diagnosis and treatment of invasive gastrointestinal fungal infection plus pulmonary infection after renal transplantation.Methods:Clinical data were analyzed retrospectively for one patient with invasive fungal infection plus pulmonary infection after renal transplantation. The middle-aged female recipient underwent allogeneic kidney transplantation due to end-stage uremia. After successful kidney transplantation, there was postprandial epigastric pain not relieved by proton pump inhibitor. Gastroscopy after admission suggested that the nature of gastric mucosal lesions was to be determined. Pathological examination and special staining confirmed mucor.Results:After clarifying her conditions, the doses of such immunosuppression as tacrolimus, mycophenolate mofetil and prednisone were tapered and discontinued when necessary and using amphotericin B liposome plus posaconazole alleviated the digestive tract symptoms. Chest tightness, fever, shortness of breath after activities hinted at pulmonary infection after renal transplantation. Treatment was guided by the results of sputum culture.Conclusions:Mucor infection is rare in digestive tract complicated with pulmonary infection after renal transplantation. Clinicians should actively search for etiological evidence, seek multidisciplinary consultations for a definite diagnosis and provide empirical anti-infection treatments. Due attention is to be paid for double infection caused by anti-infection treatments and anti-infection treatment strategy should be timely adjusted and the dosage of immunosuppressant based upon immune monitoring.

ABSTRACT

Objective:To evaluate the effect of orexin-A on programmed necrosis during cerebral ischemia-reperfusion (I/R) in rats.Methods:Thirty clean-grade healthy adult male Spraugue-Dawley rats, weighing 280-320 g, were divided into 3 groups ( n=10 each) using a random number table method: sham operation group (Sham group), cerebral I/R group (I/R group) and orexin-A group (OA group). In I/R and OA groups, a rat model of global cerebral I/R injury was established by transesophageal cardiac pacing-induced cardiac arrest and cardiopulmonary resuscitation in anesthetized animals.Orexin-A 30 μg/kg (diluted to 0.5 ml in phosphate buffer solution) was intravenously injected at 10 min before establishing the model in OA group.Phosphate buffer solution 0.5 ml was intravenously injected at 10 min before establishing the model in Sham and I/R groups.The neurological deficit score (NDS) was assessed at 24 h of reperfusion, then the rats were sacrificed, and bilateral hippocampal tissues were obtained.The morphological structure of pyramidal cells in the hippocampal CA1 region was examined after HE staining, and normal pyramidal cells were counted.Western blot was used to detect the expression of receptor-interacting protein 1 (RIP1), RIP3 and mixed-lineage kinase domain-like protein (MLKL). Immuno-histochemistry was used to count RIP1, RIP3 and MLKL positive cells in the hippocampal CA1 region.The activity of superoxide dismutase (SOD) in hippocampi was determined by xanthine oxidase method, and the content of malondialdehyde (MDA) in hippocampi was determined by thiobarbituric acid method. Results:Compared with Sham group, the normal pyramidal cell count in the hippocampal CA1 region was significantly decreased, NDS was increased, the expression of RIP1, RIP3 and MLKL protein was up-regulated, the positive cell count was increased, the content of MDA in hippocampi was increased, and the activity of SOD in hippocampi was decreased in I/R and OA groups ( P<0.05). Compared with I/R group, the count of normal pyramidal cells in the hippocampal CA1 region was significantly increased, NDS was decreased, the expression of RIP1, RIP3 and MLKL protein was down-regulated, the count of positive cells was decreased, the content of hippocampal MDA was decreased, and the activity of hippocampal SOD was increased in OA group ( P<0.05). Conclusion:The mechanism by which orexin-A reduces cerebral I/R injury may be related to inhibiting programmed necrosis in rats.

Objective@#To investigate the effect and recurrence rate of radiofrequency ablation of low temperature plasma knife in the treatment of patients with early glottal carcinoma.@*Methods@#A total of 64 cases of early glottic carcinoma in the People's Hospital of Shanxi Province from February 2012 to January 2016 were divided into observation group and control group according to the different treatment schemes, with 32 cases in each group.The observation group was treated with radiofrequency ablation of low temperature plasma knife, and the control group was performed with routine larynx cleft surgery.The surgical related indicators and postoperative complications of the two groups were recorded.The phonetic changes (amplitude perturbation, fundamental frequency perturbation and harmonic noise ratio) before and 6 months after operation of two groups were compared.After 2 years of follow-up, the recovery of vocal mucosa in the two groups after 1 year of operation and the recurrence rate of 6 months, 1 year and 2 years after operation of the two groups were statistically recorded.@*Results@#The intraoperative blood loss in the observation group was less than that in the control group(P<0.05), and the operative time and hospital stay observation time in the observation group were shorter than those in the control group(all P<0.05). The incidence rate of postoperative complications in the observation group [9.38% (3/32)] was lower than that in the control group[31.25%(10/32)], and the smoothness rate of the vocal cords at 1 year after operation in the observation group [90.63% (29/32)] was higher than that in the control group[62.50%(20/32)], the difference were statistically significant(χ2=4.730, 7.053, all P<0.05). There were no statistically significant differences in amplitude perturbation, fundamental frequency perturbation and harmonic noise ratio between the two groups(all P>0.05). After operation, the amplitude perturbation and the fundamental frequency perturbation in the observation group were lower than those of the control group, and the harmonic noise ratio was higher than that of the control group, the difference were statistically significant (t=18.618, 4.258, 2.579, all P<0.05). In the observation group, the recurrence rates at 6 months, 1 year and 2 years after operation were lower than those in the control group(χ2=4.598, 6.335, 4.730, all P<0.05).@*Conclusion@#Radiofrequency ablation of low temperature plasma knife in the treatment of patients with early glottal carcinoma can significantly reduce the intraoperative blood loss, shorten the operation time and observation time of postoperative hospital stay, and significantly reduce the incidence of postoperative complications and recurrence rate.It can also improve the patients' voice, and promote the recovery of vocal cord mucosal smooth.Besides, it has remarkable effect, and it has important significance in clinical treatment.

A patient, male, 36 years old, diagnosed as severe plaque psoriasis for 10 years and irritable bowel syndrome (IBS) for 15 years, was administrated twice FMT via both upper endoscopy and colonoscopy with a 5-week interval. The following items were used to evaluate responses: body surface area (BSA), psoriasis area and severity index (PASI), dermatology life quality index (DLQI), histological examination, intestinal symptoms, adverse reactions and serum level of tumor necrosis factor (TNF)-α. After second FMT treatment for 5 weeks, aforementioned items were improved greatly compared with those before treatment. Moreover, IBS was completely relieved and no adverse reactions were observed during the treatment and follow-up. In conclusion, FMT could be a novel therapy for psoriasis. Further clinical trials are needed to provide solid evidences.

As a member of the β-thymosin family, thymosin β 10 (Tβ10) is mainly involved in the isolation of intracellular actin. Studies have shown that the expression change of Tβ10 is closely correlated with breast cancer, esophageal cancer, liver cancer, cholangiocarcinoma, ovarian cancer and other malignant tumors. Tβ10 is expressed in different states according to the type of cancers, and the mechanism is unclear. The recent research of Tβ10 in chemotherapy and serum findings play a significant role in predicting the efficacy of chemotherapy in cancer patients.

ABSTRACT

Objective To investigate the effect and recurrence rate of radiofrequency ablation of low temperature plasma knife in the treatment of patients with early glottal carcinoma.Methods A total of 64 cases of early glottic carcinoma in the People's Hospital of Shanxi Province from February 2012 to January 2016 were divided into observation group and control group according to the different treatment schemes ,with 32 cases in each group.The observation group was treated with radiofrequency ablation of low temperature plasma knife ,and the control group was performed with routine larynx cleft surgery.The surgical related indicators and postoperative complications of the two groups were recorded.The phonetic changes (amplitude perturbation,fundamental frequency perturbation and harmonic noise ratio ) before and 6 months after operation of two groups were compared.After 2 years of follow -up,the recovery of vocal mucosa in the two groups after 1 year of operation and the recurrence rate of 6 months,1 year and 2 years after operation of the two groups were statistically recorded.Results The intraoperative blood loss in the observation group was less than that in the control group (P<0.05),and the operative time and hospital stay observation time in the observation group were shorter than those in the control group ( all P<0.05).The incidence rate of postoperative complications in the observation group [9.38%(3/32)] was lower than that in the control group [31.25%(10/32)],and the smoothness rate of the vocal cords at 1 year after operation in the observation group [90.63%(29/32)] was higher than that in the control group [62.50%(20/32)],the difference were statistically significant (χ2 =4.730,7.053,all P<0.05).There were no statistically significant differences in amplitude perturbation ,fundamental frequency perturbation and harmonic noise ratio between the two groups (all P>0.05).After operation,the amplitude perturbation and the fundamental frequency perturbation in the observation group were lower than those of the control group,and the harmonic noise ratio was higher than that of the control group , the difference were statistically significant (t=18.618,4.258,2.579,all P<0.05).In the observation group,the recurrence rates at 6 months, 1 year and 2 years after operation were lower than those in the control group ( χ2 =4.598,6.335,4.730,all P<0.05).Conclusion Radiofrequency ablation of low temperature plasma knife in the treatment of patients with early glottal carcinoma can significantly reduce the intraoperative blood loss ,shorten the operation time and observation time of postoperative hospital stay ,and significantly reduce the incidence of postoperative complications and recurrence rate.It can also improve the patients'voice,and promote the recovery of vocal cord mucosal smooth.Besides,it has remarkable effect,and it has important significance in clinical treatment.

Objective To prepare 3D printed porous tracheal graft fabricated by PCL and to select the appropriate pore size and surface modification techniques,in order to explore its effect on cell behavior.Methods The PCL porous tracheal graft was prepared by 3D printing technology and biomechanical properties of the graft were measured by means of longitudinal tension,radial compression and three-point bending test.The porous grafts were surface-modified through hydrolysis,amination and nanocrystallization treatment and then characterized by energy dispersive spectroscopy(EDS).The effect of different pore sizes and surface modifications on the cell proliferation behavior was evaluated by CCK-8 and scanning electron microscopy (SEM).Results The 3 D printed porous tracheal graft had similar morphology with the native tracheas(P > 0.05) and better biomechanical properties(P <0.05).It was more suitable for cell adhesion and proliferation when the pore size is 200 μm (P < 0.05).Compared to hydrolysis and amination,nanocrystallization treatment successfully improved the cytotropism of the 3D printed tracheal graft(P < 0.05).Conclusion 3 D printed porous tracheal graft shows favorable biomechanical properties.The appropriate pore size of the 3D printed porous tracheal graft is 200 μm and the appropriate surface modification techniques is nanocrystallization.

ABSTRACT

Objective To evaluate the role of nuclear factor erythroid 2-related factor 2 ( Nrf2 ) signaling pathway in endoplasmic reticulum stress response during lipopolysaccharide ( LPS)-induced acute lung injury ( ALI) in mice. Methods Forty clean-grade healthy male C57BL∕6 mice, aged 6-8 weeks, weighing 22-26 g, were divided into 4 groups ( n=10 each) using a random number table method: control group ( group C) , group ALI, salubrinal group ( group S) and salubrinal plus brusatol group ( group S+B) . Animals were intratracheally instilled with 5 mg∕kg of LPS diluted in normal saline to establish the model of ALI. Animals were intratracheally instilled with 100 μl of normal saline in group C. Mice in group S were intraperitoneally injected with endoplasmic reticulum stress response inhibitor 1 mg∕kg salubrinal at 1 and 24 h after LPS instillation. Mice of group S+B were intraperitoneally injected with brusatol 2 mg∕kg once every other day for 10 days prior to LPS instillation, and the other treatments were similar to those previously de-scribed in group S. Mice were sacrificed at 48 h after LPS administration, and lungs were removed for mi-croscopic examination of the pathological changes of lung tissues which were scored and for determination of contents of IL-17A, tumor necrosis factor-alpha ( TNF-α) and interleukin-6 ( IL-6) in lung tissues ( by en-zyme-linked immunosorbent assay) and expression of Nrf2, CCAAT∕enhancer-binding protein homologous protein (CHOP) and caspase-12 in lung tissues (by Western blot). Lung water content was calculated. Results Compared with group C, the lung water content and contents of IL-17A, TNF-α and IL-6 were significantly increased, the expression of CHOP and caspase-12 in cytoplasma was up-regulated, and the ex-pression of Nrf2 in nuclei was down-regulated in ALI and S+B groups, and the lung water content and con-tents of IL-17A, TNF-α and IL-6 were significantly increased, the expression of Nrf2 in nuclei and CHOP in cytoplasma was up-regulated, and the expression of caspase-12 was down-regulated in group S ( P<0. 05) . Compared with group ALI, the lung water content and contents of IL-17A, TNF-α and IL-6 were significantly decreased, the expression of CHOP and caspase-12 in cytoplasma was down-regulated, the ex-pression of Nrf2 in nuclei was up-regulated ( P<0. 05) , and the pathological changes were significantly at-tenuated in group S. Compared with group S, the lung water content and contents of IL-17A, TNF-α and IL-6 were significantly increased, the expression of CHOP and caspase-12 in cytoplasma was up-regulated, the expression of Nrf2 in nuclei was down-regulated ( P<0. 05) , and the pathological changes of lung tis-sues were accentuated in group S+B. Conclusion Nrf2 signaling pathway is involved in the process of en-doplasmic reticulum stress response during LPS-induced ALI in mice.
